Status Public on Sep 15, 2022
Title hPSC cardiomyocytes at Day 15 of differentiation (D15_F05)_gene
Sample type RNA

Source name hPSC-derived cardiomyocytes
Organism Homo sapiens
Characteristics cell type: hPSC-derived cardiomyocytes
age: d15
cell lineage: Coriell Cell Repositories (AICS-0037-172)
Biomaterial provider Coriell Cell Repositories (AICS-0037-172)
https://www.coriell.org/0/Sections/Search/Sample_Detail.aspx?Ref=AICS-0037-172&PgId=166
Treatment protocol Cardiac cell differentiation protocol followed Burridge et al., 2014 specifications. No treatment was applied after cell differentiation.
Growth protocol Prior to cardiac differentiation hPSC cells were maintained in E8:mTeSR [(1:1), Stemcell Technologies, #85850] with daily change. Established hPSC-CM were cultivated with RPMI-1640 supplemented with B27 and insulin until the end of protocol.
Extracted molecule total RNA
Extraction protocol We used the RNeasy Micro Kit (Qiagen) for total RNA isolation
Label biotin
Label protocol Samples were enzymatically fragmented and biotinylated using RNA samples for whole transcriptome expression analysis with GeneChipTM Whole Transcript (WT) Expression Arrays (appliedbiosystems)

Hybridization protocol Samples were hybridized with GeneChipTM WT PLUS Reagent Kit (Affymetrix) and scanned at GeneTitan ™ Microarray System
Scan protocol The entire protocol for processing, running and scanning the samples followed the manufacturer's instruction [GeneChip ™ WT PLUS Reagent Kit Manual] at GeneTitan ™ Microarray System
Data processing The .CEL files were analyzed using the Transcriptome Analysis Console software (TAC v4.0) from AppliedBiosystems
