Title |
plant_treated_with_35°C_heat_rep2 |
Sample type |
RNA |
Source name |
20d of above-ground parts, Arabidopsis thaliana, 35°C heat, 4hrs, replicate2
Organism |
Arabidopsis thaliana |
Characteristics |
ecotype: Col-0 tissue: above-ground parts age: 20d genotype: wild type treatment: 35°C heat, 4hrs
Treatment protocol |
Soil-grown plants were treated with 150mM, 300mM of salt and/or 200mM, 400mM of mannitol for 16 hrs by feeding the solution and/or 35°C heat for 4 hrs by placing plants in incubator.
Growth protocol |
All plants were grown in MS (1% sucrose) agar media for first 10 days and in soil for next 10 days in 16hr light / 8hr dark cycle in 23 Celsius degree before stress treatment
Extracted molecule |
total RNA |
Extraction protocol |
Plant RNeasy Mini kit (QIAGEN) was used to isolate total RNA.
Label |
Cy3
Label protocol |
1 µg of total RNA was used for labeling procedure using QickAmp Labeling kit (One-color) (Agilent).
Hybridization protocol |
Hybridized using Gene Expression Hybridization kit (Agilent) and washed by Gene Expression wash pack (Agilent).
Scan protocol |
Scanned on an Agilent G2505B scanner.
Description |
Gene expression of plants treated with 35°C heat
Data processing |
Images were quantified using Agilent Feature Extraction Software (version 9.1.3.1). Agilent Feature Extraction Software (v 9.1.3.1) was used for background subtraction. After that, signal of each probe was divided by median value of filter-passed probes.
