Cells were grown for 1 days on irradiated feeders after trypsinisation and either cultured for 1 more day under these conditions or induced for 24h by the addition of DOX.

Cells were passaged according to standard procedures with standard ES medium (Non essential AA, Pyruvate, PSG, LIF, 15% serum, beta-mercaptoethanol)

After 2 days of culture, cells were trypsinized, resuspended in 15ml of medium, allowed to settle for 10 minutes to eliminate feeders. The top 50% of the suspension was used for RNA preparation/Trizol.
