Mice were sub-lethally irradiated for 2 fractions of 310cGy at 10 days of age, then PTH administration began 24 hours later for 5 days.

Extraction protocol |
Long bones were harvested and flushed with Trizol. Extraction of total RNA was performed according to the manufacturer's instructions. RNA was cleaned up using the the Qiagen RNEasy Kit.

Biotinylated cDNA were prepared according to the Nugen Applause protocol from 100 ng total RNA (Nugen Applause WT-AMP ST V1 Manual, version 29 April 2009, NuGEN).

Hybridization protocol |
Following Labeling, 4 ug of cDNA were hybridized for 16 hr at 45C on Mouse gene ST 1.0 Arrays. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

Scan protocol |
GeneChips were scanned using the Affymetrix 3000 7G GeneChip Scanner with Autoloader.

Data processing |
Data was analyzed using the limma and affy Bioconductor packages implemented in the R-statistical environment. RMA was used to normalize the data and fit log2 transformed expression values.
