Decreased mitochondrial-related gene expression in adipose tissue after acute sprint exercise in humans

(Submitter supplied) The aim was to examine the acute effects of sprint exercise (SIT) on global gene expression in subcutaneous adipose tissue (AT) in healthy subjects to improve the understanding of how SIT may contribute to the maintenance of a healthy body weight. An SIT-induced upregulation of genes related to mitochondrial and fat metabolism was hypothesized. A total of 15 subjects performed three 30-s all-out sprints (SIT). more...

Molecular subtypes are associated with clinical benefit in cisplatin-treated metastatic urothelial cancer patients

(Submitter supplied) Cisplatin-based combination chemotherapy (CHT) is standard of care in locally advanced and metastatic urothelial cancer (mUC). No predictive biological biomarkers are available for clinical use. Here, we investigate the impact of molecular subtypes in relation to response and survival in mUC patients treated with first-line CHT. Molecular subtype classification according to Lund taxonomy was performed by tumour transcriptomic profiling and immunostaining in a retrospective cohort with mUC patients treated with CHT. more...

Microarray analysis of human cord blood derived mast cells (hCBMC) activated by Interleukin-33 (IL-33)

(Submitter supplied) Mast cells respond to acute and chronic exposure to IL-33 by differing the expression of their receptors, cytokines and proteases. We aim to characterize the distinct acute and chronic responses of mast cells to IL-33 by analyzing hCBMC expression profile of pro- and anti- inflammatory mediators.

Expression data from AGS cells with Helicobacter pylori infection

(Submitter supplied) Helicobacter pylori (H. pylori) is a human pathogen that infects almost half of the world’s population. Infection with H. pylori is frequently associated with chronic gastritis and can even lead to gastric and duodenal ulcers and gastric cancer. Although the persistent colonization of H. pylori and the development of H. pylori-associated gastritis remain poorly understood, it is believed that, in gastric mucosa, the modulated gastric epithelial cells (GECs) by H. more...

A role for miR-4734 in conditional suppression of ER stress-associated pro-inflammatory responses [starved vs control HFF]

(Submitter supplied) Prolonged metabolic stress can lead to severe pathologies. In metabolically challenged primary fibroblasts, we assigned a novel role for the poorly characterized miR-4734 in restricting ATF4 and IRE1-mediated upregulation of a set of pro-inflammatory cytokines and endoplasmic reticulum (ER) stress-associated genes. Conversely, inhibition of this miRNA augmented the expression of those genes. Mechanistically, miR-4734 was found to restrict the expression of NFKBIZ, that is required for optimal expression of the pro-inflammatory genes and whose mRNA is targeted directly by miR-4734. more...

Effect of miR-122 overexpression of mRNA distribution on the polyribosomes

(Submitter supplied) The aim of this study was to identify at genome-wide level, the effect of miR-122 on the distribution of its responsive genes on the polyribosomes. For this purpose human hepatoma cells Huh-7 were transfected for 48 hrs with LNA-based miR-122 inhibitors (122-PM) or miR-122 mimics (122-MIM). Following the treatment with cycloheximide, cells were lysed and layered on a 10-50% sucrose gradient. Following ultracetrifigation the polyribosomes were separated in 12 different fractions

Treatment with HKMTI-1-005, a dual inhibitor of EZH2 and G9a/GLP histone methyltransferases, promotes differentiation of acute myeloid leukaemialeukemia

(Submitter supplied) The canonical function of EZH2 is methylation of H3K27, although important non-canonical roles have recently been described. EZH2 mutation or deregulated expression has been conclusively demonstrated in the pathogenesis and response to treatment of acute myeloid leukemia (AML), thus making it an attractive therapeutic target. In this study, we therefore investigated whether inhibition of EZH2 might also improve the response of non-APL AML cells to ATRA-based therapy. more...

NPC stem-like cells [Affymetrix]

(Submitter supplied) Stem-like cells of nasopharyngeal cancer were enriched by 3 different methods: 1) Side population (SP) assay, 2) LMP2A over-expression and 3) spheroid culture. SP cells and LMP2A over-expressed cells were inoculated into immunocompromised mice for xenograft tumors. Transcriptomes of all 5 of these in vitro and in vivo generated NPC stem-like cells were then analyzed and compared to their counterparts by microarray. more...
