Thyroid hormone-responsive genes in primary cultures of rat hepatic cells

(Submitter supplied) cDNA microarray analysis was applied to primary cultures of rat hepatic cells treated with triiodothyronine (T3) at 10-9 M for 24 hrs to identify the differentially expressed genes. A limited number of genes were listed, and only 3 of them, pyridoxal kinase (Pdxk), phosphoe-nolpyruvate carboxykinase 1 (Pck1), and solute carrier family 17 member 2 (Slc17a2), were con-firmed to be up-regulated by quantitative RT-PCR.

Identifying bone marrow microenvironmental alterations in multiple myeloma and its precursor conditions using whole bone marrow biopsies

(Submitter supplied) Tumor immune microenvironmental alterations occur early in multiple myeloma (MM) development. In this study, we aim to systematically characterize the tumor immune microenvironment (TME) and the tumor-immune interactions from precursor stages, i.e., monoclonal gammopathy of undetermined significance (MGUS) and smoldering MM (SMM), to newly diagnosed MM, comparing these to healthy donors. (Submitter supplied) C-type lectin dectin-1 is known to recognize beta-glucans such as curdlan and propagate intracellular signaling for activation of NF-kappa B to induce anti-fungal immunity. Recently, we found that adaptor protein 3BP2 is required for the dectin-1-induced activation of NF-kappa B. Combined inhibition of histone methyltransferases EZH2 and DOT1L is an effective therapy for neuroblastoma

(Submitter supplied) The child cancer, neuroblastoma (NB), is characterised by a low incidence of mutations and strong oncogenic embryonal driver signals. Many new targeted epigenetic modifier drugs have failed in human trials as monotherapy. We performed a high‐throughput, combination chromatin‐modifier drug screen against NB cells. We screened 13 drug candidates in 78 unique combinations. We found that the combination of two histone methyltransferase (HMT) inhibitors: GSK343, targeting EZH2, and SGC0946, targeting DOT1L, demonstrated the strongest synergy across 8 NB cell lines, with low normal fibroblast toxicity. more...

Pseudohypoxia Due to PHD2 Deficiency Induces Endothelial Resistance to Apoptosis and Inflammation Independent of HIF2 alpha via AKT Activation and AIP1 Loss

Regulatory T cells crosstalk with tumor and endothelium through lymphotoxin signaling [array]

Tregs use the surface LTa1b2 to preferentially stimulate LTbR nonclassical NFkB signaling on both tumor and LECs to accelerate tumor growth and metastasis.
